    High level triggers for explosive mafic volcanism: Albano Maar, Italy

    Colli Albani is a quiescent caldera complex located within the Roman Magmatic Province (RMP), Italy. The recent Via dei Laghi phreatomagmatic eruptions led to the formation of nested maars. Albano Maar is the largest and has erupted seven times between ca 69-33ka. The highly explosive nature of the Albano Maar eruptions is at odds with the predominant relatively mafic (SiO2=48-52wt.%) foiditic (K2O=9wt.%) composition of the magma. The deposits have been previously interpreted as phreatomagmatic, however they contain large amounts (up to 30%vol) of deep seated xenoliths, skarns and all pre-volcanic subsurface units. All of the xenoliths have been excavated from depths of up to 6km, rather than being limited to the depth at which magma and water interaction is likely to have occurred, suggesting an alternative trigger for eruption. High precision geochemical glass and mineral data of fresh juvenile (magmatic) clasts from the small volume explosive deposits indicate that the magmas have evolved along one of two evolutionary paths towards foidite or phonolite. The foiditic melts record ca. 50% mixing between the most primitive magma and Ca-rich melt, late stage prior to eruption. A major result of our study is finding that the generation of Ca-rich melts via assimilation of limestone, may provide storage for significant amounts of CO2 that can be released during a mixing event with silicate magma. Differences in melt evolution are inferred as having been controlled by variations in storage conditions: residence time and magma volume. Ā© 2013

    Lake Ohridā€™s tephrochronological dataset reveals 1.36ā€‰Ma of Mediterranean explosive volcanic activity

    Tephrochronology relies on the availability of the stratigraphical, geochemical and geochronological datasets of volcanic deposits, three preconditions which are both often only fragmentary accessible. This study presents the tephrochronological dataset from the Lake Ohrid (Balkans) sediment succession continuously reaching back to 1.36ā€‰Ma. 57 tephra layers were investigated for their morphological appearance, geochemical fingerprint, and (chrono-)stratigraphic position. Glass fragments of tephra layers were analyzed for their major element composition using Energy-Dispersive-Spectroscopy and Wavelength-Dispersive Spectroscopy and for their trace element composition by Laser Ablation-Inductively Coupled Plasma-Mass Spectrometry. Radiometric dated equivalents of 16 tephra layers and orbital tuning of geochemical proxy data provided the basis for the age-depth model of the Lake Ohrid sediment succession. The age-depth model, in turn, provides ages for unknown or undated tephra layers. This dataset forms the basis for a regional stratigraphic framework and provides insights into the central Mediterranean explosive volcanic activity during the last 1.36ā€‰Ma

    No evidence for tephra in Greenland from the historic eruption of Vesuvius in 79 CE: implications for geochronology and paleoclimatology

    Volcanic fallout in polar ice sheets provide important opportunities to date and correlate ice-core records as well as to investigate the environmental impacts of eruptions. Only the geochemical characterization of volcanic ash (tephra) embedded in the ice strata can confirm the source of the eruption, however, and is a requisite if historical eruption ages are to be used as valid chronological checks on annual ice layer counting. Here we report the investigation of ash particles in a Greenland ice core that are associated with a volcanic sulfuric acid layer previously attributed to the 79 CE eruption of Vesuvius. Major and trace element composition of the particles indicates that the tephra does not derive from Vesuvius but most likely originates from an unidentified eruption in the Aleutian arc. Using ash dispersal modelling, we find that only an eruption large enough to include stratospheric injection is likely to account for the sizeable (24ā€“85 Ī¼m) ash particles observed in the Greenland ice at this time. Despite its likely explosivity, this event does not appear to have triggered significant climate perturbations, unlike some other large extra-tropical eruptions. In light of a recent re-evaluation of the Greenland ice-core chronologies, our findings further challenge the previous assignation of this volcanic event to 79 CE. We highlight the need for the revised Common Era ice-core chronology to be formally accepted by the wider ice-core and climate modelling communities in order to ensure robust age linkages to precisely dated historical and paleoclimate proxy records

    Severe polyposis in Apc1322T mice is associated with submaximal Wnt signalling and increased expression of the stem cell marker Lgr5

    Background and aims: Adenomatous polyposis coli (APC) is a tumour suppressor gene mutated in the germline of patients with familial adenomatous polyposis (FAP) and somatically in most colorectal cancers. APC mutations impair Ī²-catenin degradation, resulting in increased Wnt signalling. The most frequent APC mutation is a codon 1309 truncation that is associated with severe FAP. A previous study compared two mouse models of intestinal tumorigenesis, ApcR850X (Min) and. Apc1322T (1322T), the latter a model of human codon 1309 changes. 1322T mice had more severe polyposis but, surprisingly, these tumours had lower levels of nuclear Ī²-catenin than Min tumours. The consequences of these different Ī²-catenin levels were investigated. Methods: Enterocytes were isolated from 1322T and Min tumours by microdissection and gene expression profiling was performed. Differentially expressed Wnt targets and other stem cell markers were validated using quantitative PCR, in situ hybridisation and immunohistochemistry. Results: As expected, lower nuclear Ī²-catenin levels in 1322T lesions were associated with generally lower levels of Wnt target expression. However, expression of the Wnt target and stem cell marker Lgr5 was significantly higher in 1322T tumours than in Min tumours. Other stem cell markers (Musashi1, Bmi1 and the Wnt target Cd44) were also at higher levels in 1322T tumours. In addition, expression of the Bmp antagonist Gremlin1 was higher in 1322T tumours, together with lower Bmp2 and Bmp4 expression. Conclusions: The severe phenotype caused by truncation of Apc at codon 1322 is associated with an increased number of stem cells. Thus, a submaximal level of Wnt signalling favours the stem cell phenotype and this may promote tumorigenesis. A level of Wnt signalling exists that is too high for optimal tumour growth

    The Healthy Lifestyles Programme (HeLP), a novel school-based intervention to prevent obesity in school children: study protocol for a randomised controlled trial

    Background: Over the last three decades there has been a substantial increase in the proportion of children who are overweight or obese. The Healthy Lifestyles Programme (HeLP) is a novel school-based intervention, using highly interactive and creative delivery methods to prevent obesity in children. Methods/Design: We describe a cluster randomised controlled trial to evaluate the effectiveness and cost effectiveness of HeLP. The intervention has been developed using intervention mapping (involving extensive stakeholder involvement) and has been guided by the Information, Motivation, Behavioural Skills model. HeLP includes creating a receptive environment, drama activities, goal setting and reinforcement activities and runs over three school terms. Piloting showed that 9 to 10 year olds were the most receptive and participative. This study aims to recruit 1,300 children from 32 schools (over half of which will have ā‰„19% of pupils eligible for free school meals) from the southwest of England. Participating schools will be randomised to intervention or control groups with baseline measures taken prior to randomisation. The primary outcome is change in body mass index standard deviation score (BMI SDS) at 24 months post baseline. Secondary outcomes include, waist circumference and percent body fat SDS and proportion of children classified as overweight or obese at 18 and 24 months and objectively measured physical activity and food intake at 18 months. Between-group comparisons will be made using random effects regression analysis taking into account the hierarchical nature of the study design. An economic evaluation will estimate the incremental cost-effectiveness of HeLP, compared to control, from the perspective of the National Health Service (NHS)/third party payer. An in-depth process evaluation will provide insight into how HeLP works, and whether there is any differential uptake or engagement with the programme. Discussion: The results of the trial will provide evidence on the effectiveness and cost effectiveness of the Healthy Lifestyles Programme in affecting the weight status of children

    Improving the efficiency of aerosolized insecticide testing against mosquitoes

    Developing robust and standardised approaches for testing mosquito populations against insecticides is vital for understanding the effectiveness of new active ingredients or formulations. Methods for testing mosquito susceptibility against contact insecticides or products, such as those delivered through public health programmes, are well-established and standardised. Nevertheless, approaches for testing volatile or aerosolized insecticides used in household products can be challenging to implement efficiently. We adapted WHO guidelines for household insecticides to develop a standardised and higher-throughput methodology for testing aerosolized products in a Peet Grady test chamber (PG-chamber) using caged mosquitoes and an efficient decontamination method. The new approach was validated using insecticide resistant and susceptible Aedes and Anopheles mosquito colonies. An added feature is the inclusion of cage-facing cameras to allow real-time quantification of knockdown following insecticide exposure. The wipe-based decontamination method was highly effective for removing pyrethroids' aerosolized oil-based residues from chamber surfaces, withā€‰<ā€‰2% mortality recorded for susceptible mosquitoes tested directly on the surfaces. There was no spatial heterogeneity for knockdown or mortality of caged mosquitoes within the PG chamber. The dual-cage approach we implement yields eight-times the throughput compared to a free-flight protocol, allows simultaneous testing of different mosquito strains and effectively discriminates susceptible and resistant mosquito colonies tested side-by-side

    Trace element and Pb isotope fingerprinting of atmospheric pollution sources: A case study from the east coast of Ireland

    Unravelling inputs of multiple air pollution sources and reconstructing their historic contribution can be a difficult task. Here, new trace metal concentrations and Pb isotope data were combined for a radionuclide (210Pb-241Am) dated peat core from the Liffey Head bog (LHB) in eastern Ireland in order to reconstruct how different sources contributed to the atmospheric pollution over the past century. Highest enrichments in the heavy metals Pb, Cu, Ag, Sn, and Sb, together with a Pb isotope composition (206Pb/204Pb: 18.351 Ā± 0.013; 206Pb/207Pb: 1.174 Ā± 0.012) close to that of the Wicklow mineralisation demonstrates significant aerial influx of heavy metals from local mining and smelting activities during the 19th century until ca. 1940s. A dramatic compositional shift defined by elevated Co, Cr, Ni, Mo, Zn, and V enrichments and a sharp transition towards unradiogenic 206Pb values (206Pb/204Pb: 18.271 Ā± 0.013ā€“17.678 Ā± 0.006; 206Pb/207Pb: 1.170 Ā± 0.012ā€“1.135 Ā± 0.007) is documented from the 1940s until ca. 2000. These are attributed to the atmospheric impact of fossil fuels and especially leaded petrol, modelled to have contributed between 6 and 78% to the total Pb pollution at this site. The subsequent turn to a more radiogenic Pb isotope signature since 2000 in Ireland is clearly documented in the investigated archive (206Pb/204Pb: 17.930 Ā± 0.006; 206Pb/207Pb: 1.148 Ā± 0.007) and reflects the abolishment of leaded petrol. However, there remains a persisting and even increasing pollution in Ni, Mo, Cu, and especially Zn, collectively originating from countrywide use of fossil fuels(peat, coal, heating oil, and unleaded vehicle fuels) for domestic and industrial purposes. This illustrates the continued anthropogenic influence on important natural archives such as bogs in Ireland despite the phase-out of leaded petrol
